Introduction

The discovery of chitinolytic bacteria from hot springs has opened up new avenues for research in the field of biotechnology. Chitinase enzymes, produced by these bacteria, play a crucial role in the decomposition of chitin, a polymer found in fungi cell walls and arthropod exoskeletons. In this study, we aimed to screen chitinolytic bacteria from the hot water source of Penen, Sibiru-Biru District, Deli Serdang Regency, and characterize its chitinase enzymes.

Research Methodology

To isolate chitinolytic bacteria, we used a medium with minimal salt content, where colloidal chitin served as the sole carbon source. The incubation process lasted for 7 days at 53 ° C. To evaluate the chitinase index, we observed the clear zone produced by each isolate on the same medium. The bacterial isolation process was carried out using a medium with minimal salt content, where colloidal chitin functions as the only carbon source.

Isolation and Characterization of Chitinolytic Bacteria

The bacterial isolation process resulted in the isolation of ten chitinolytic bacterial isolates. Among these, JP2 isolates showed the highest ability to utilize chitin, with a chitinase index of 1.65. The highest chitinase production of JP2 isolates was found on the 4th day, with measurable chitinase activity of 0.0063 U/ml. The highest activity of chitinase was also recorded at the concentration of ammonium sulfate 50%, which is 0.0087 U/ml, with specific activity reaching 0.046 U/mg.

Results and Discussion

The results showed that chitinase activity increased significantly at the optimum pH 6 and the optimum temperature 60 ° C, where chitinase activity reached 0.05066 U/ml with specific activity of 3,8113 U/mg after the dialysis process. Further analysis shows the values of KM and VMAX each are 0.321 mg/ml and 71.429 μg. Low Km values indicate that the chitinase enzymes from JP2 isolates have high affinity to the chitin substrate, which means it can work efficiently even in low substrate concentrations.

Identification of Bacteria

Identification of bacteria was carried out through the analysis of the 16s RRNA gene, which showed that JP2 isolates are classified into the species of Bacillus licheniformis. This type of bacteria is known to have the ability to produce chitinase enzymes, which play an important role in the process of decomposing chitin.

Conclusion

This study succeeded in isolating chitinolytic bacteria from hot springs and chitinase characterization. JP2 isolates from Bacillus licheniforms show excellent potential in chitinase activity that can be useful in biotechnology applications, especially in the organic waste treatment industry and enzyme production. A deeper understanding of the characteristics of this enzyme can open new opportunities in developing environmentally friendly and efficient bioproces.
